Resident Evil Zero

Of all the games Wesker has been in, his role in Resident Evil Zero was the smallest, but despite this fact, it probably had the biggest impact on both the series as a whole, and on his career. The events that are uncovered in this game set into motion that which eventually caused him to become a Tyrant, and with that said, here's the explanation.

When the Ecliptic Express crashes in the Arklay Forest, a train which carries Umbrella employees from Raccoon City into the forest to work on a daily basis, the S.T.A.R.S. Bravo Team gets caught up in the middle of it. Rookie member Rebecca Chambers is twisted into one of the deepest secrets of Umbrella's business, and has to put her trust into a complete stranger, Billy Coen, to get out alive.

What the two don't realize at this point in time is that the cause of the ravenous monsters inhabiting the train is a viral outbreak, one scattered by a mysterious man. Wesker meets with his old colleague, Dr. Birkin, about this chain of events, and even he is in the dark about how the T Virus made an outbreak to begin with. But he knows one thing for certain, that it's not time for questions, and tells Birkin that their only choice is to destroy the train completely, using his Umbrella Agents to get the train to operate once again and bring it back to the nearest branchline.

However, due to the mysterious mans interference, the train crashes into the old training facility of Umbrella, and it's not too long after that when the man reveals why. In an interrupted broadcast, he reveals himself to both Wesker and Birkin, that he is actually the late Dr. James Marcus, whom had been assassinated ten years before by Umbrella, and more specifically, with the help of Wesker and Birkin. He scattered the T Virus in order to get revenge on his old wards and on Umbrella.

With the revelation of this knowledge, Wesker knows it's time to put his own plans into motion. If news of the outbreaks escapes, not only will his and Birkin's career be over, but so will Spencer's, and Umbrella will be finished. So in light of this knowledge, he informs Birkin that he will "Simply say goodbye to Umbrella." With the T Virus having been completed, and their only remaining task to gather combat data, he'll use his S.T.A.R.S. Alpha Team as the guinea pigs he'd created them for, and lure them into the Spencer Mansion not too far away from the Training Facility where the virus has also made an outbreak.

This is the extent of Wesker's cameo in this game, the only other time we see him or Birkin being in a flashback by Dr. Marcus of when he was assassinated. We find out that after his death, T Virus infected leeches manifested themselves inside his body, and though it took them years, they eventually brought him back to life, and he then sought out his revenge. However, due to Rebecca's and Billy's efforts, Dr. Marcus died before he was able to see his plans through, preventing him from revealing anything more to anyone than was necessary, and also allowing Wesker the time he needed to try and pull his own plans off.
